Validasi kit elisa komersial untuk pengukuran hormon progesteron pada gajah sumatra abstrak gajah sumatra (elephas maximus sumatranus) merupakan satwa liar berstatus terancam punah dan tercantum dalam appendix i cites. monitoring hormon progesteron berperan penting dalam penentuan status reproduksi seperti estrus, kebuntingan, dan postpartus. namun, kit elisa yang divalidasi khusus untuk spesies ini belum tersedia. oleh karena itu, diperlukan validasi kit elisa komersial sebelum digunakan untuk tujuan reproduksi gajah. penelitian ini bertujuan untuk mengevaluasi validitas analitik dan biologis kit elisa progesteron komersial (eia-1561) untuk pengukuran hormon progesteron pada darah gajah sumatra. validasi analitik dilakukan melalui uji parallelism, akurasi, dan presisi (intra-assay dan inter-assay), sedangkan validasi biologis dilakukan dengan membandingkan kadar progesteron antara gajah prepartus dan postpartus. hasil uji parallelism menunjukkan kurva standar dan kurva sampel sejajar dengan r = 0,94 (p = 0,001). uji akurasi adalah 101%. uji presisi menghasilkan %cv intra-assay sebesar 3,18% (low quality control) dan 5,25% (high quality control), serta %cv inter-assay sebesar 0,89% (low quality control) dan 3,79% (high quality control). disimpulkan bahwa kit elisa eia-1561 memiliki validitas analitik yang baik dan berpotensi digunakan untuk analisis progesteron pada gajah sumatra. kata kunci: gajah sumatra, kit elisa, progesteron, validasi

Abstract
VALIDATION OF COMMERCIAL ELISA KITS FOR MEASURING THE PROGESTERONE HORMONE IN SUMATRAN ELEPHANTS ABSTRACT The sumatran elephant (Elephas maximus sumatranus) is classified as an endangered species and is listed in Appendix I of CITES. Monitoring progesterone levels plays a crucial role in determining reproductive status such as estrus, prepartus and postpartus conditions. However, ELISA kits specifically validated for this species are not yet available in Indonesia. Therefore, validation of commercial ELISA kits is essential before they can be applied for reproductive monitoring in elephants. This study aimed to evaluate the analytical and biological validity of a commercial progesterone ELISA kit (EIA-1561) for measuring progesterone levels in sumatran elephant blood. Analytical validation included tests of parallelism, accuracy, and precision (intra- assay and inter-assay), while biological validation was conducted by comparing progesterone concentrations between pregnant and postpartus elephants. The parallelism test showed that the standard and sample curves were parallel, with a correlation coefficient of r = 0.94 (p = 0.001). The accuracy test showed a recovery rate of 101%. Precision testing resulted in intra-assay %CVs of 3.18% (low quality control) and 5.25% (high quality control), and inter-assay %CVs of 0.89% (low quality control) and 3.79% (high quality control). It is concluded that the EIA-1561 ELISA kit demonstrates good analytical validity and has the potential to be used for progesterone analysis in sumatran elephants.
