Kulit limbah buah nanas dan pepaya serta limbah ikan dijadikan bahan baku dalam pembuatan probiotik ragi dan bakteri asam laktat (rabal) dengan tujuan mengolah sisa-sisa bahan menjadi produk suplemen ternak yang bernilai guna, sekaligus mengurangi limbah dari proses pengolahan keumamah yang tidak dikonsumsi. penelitian ini bertujuan untuk mengetahui efek penambahan limbah ikan pada probiotik ragi dan bakteri asam laktat (rabal) terhadap aktivitas antibakteri. rancangan penelitian yang digunakan berupa rancangan acak lengkap (ral) faktorial, yang terdiri dari enam perlakuan yaitu p0 yaitu tanpa penambahan limbah ikan (0%), p1 yaitu penambahan limbah ikan 10%, p2 yaitu penambahan limbah ikan 20%, p3 penambahan limbah ikan 30% serta kontrol positif (k(+)) yaitu tetrasiklin dan kontrol negatif (k(-)) yaitu aquades dengan hari inkubasi probiotik rabal selama 0 jam (h0), 24 jam (h1), 72 jam (h3), 120 jam (h5), 168 jam (h7). uji aktivitas antibakteri dilakukan dengan metode difusi cakram atau kirby-bauer. hasil penelitian didapatkan bahwa aktivitas antibakteri pada probiotik rabal dilihat berdasarkan rata-rata diameter zona hambat dengan bakteri indikator escherichia coli dan staphylococcus aureus didapatkan rata-rata zona hambat terbesar yaitu pada probiotik rabal ditambahkan 10% limbah ikan (p1) dengan masa inkubasi 168 jam (h7) yaitu 2,23 mm dan 2,73 mm. berdasarkan hasil penelitian dapat disimpulkan bahwa probiotik rabal yang ditambah 10% limbah ikan (p1) dan diinkubasi selama 168 jam (h7) memiliki aktivitas antibakteri terbesar dibandingkan dengan perlakuan lainnya.



Abstract

Pineapple and papaya fruit waste skin and fish waste are used as raw materials in the manufacture of probiotic yeast and lactic acid bacteria (RABAL) with the aim of processing the remaining ingredients into valuable livestock supplement products, while reducing waste from the processing of unconsumed keumamah. This study aims to determine the effect of adding fish waste to probiotic yeast and lactic acid bacteria (RABAL) on antibacterial activity. The research design used was a factorial Completely Randomised Design (CRD), consisting of six treatments, namely P0, namely without the addition of fish waste (0%), P1, namely the addition of 10% fish waste, P2, namely the addition of 20% fish waste, P3, the addition of 30% fish waste and positive control (K (+)), namely tetracycline and negative control (K (-)), namely distilled water with incubation days of RABAL probiotics for 0 hours (H0), 24 hours (H1), 72 hours (H3), 120 hours (H5), 168 hours (H7). Antibacterial activity test was conducted by disc diffusion or Kirby-Bauer method. The results showed that the antibacterial activity of RABAL probiotics was seen based on the average diameter of the inhibition zone with the indicator bacteria Escherichia coli and Staphylococcus aureus, the largest average inhibition zone was obtained in RABAL probiotics added 10% fish waste (P1) with an incubation period of 168 hours (H7), namely 2.23 mm and 2.73 mm. Based on the results of the study, it can be concluded that RABAL probiotics added with 10% fish waste (P1) and incubated for 168 hours (H7) have the greatest antibacterial activity compared to other treatments.
