Telah dilakukan penelitian pengaruh kombinasi dari jenis zat pengatur tumbuh auksin naphthalene acetic acid (naa) dengan sitokinin benzylaminopurin (bap) terhadap eksplan tunas aksilar dan daun cempaka kuning (magnolia champaca l.) secara in vitro. penelitian ini dilaksanakan sejak bulan januari hingga oktober 2024 di laboratorium fisiologi (bagian tumbuhan) departemen biologi fakultas matematika dan ilmu pengetahuan alam universitas syiah kuala. penelitian ini menggunakan rancangan acak lengkap (ral) dua faktor, yaitu konsentrasi naa dan konsentrasi bap. perlakuan naa dan bap yang terdiri dari 36 perlakuan dan setiap kombinasi perlakuan diulang sebanyak tiga kali. perlakuan yang digunakan yaitu penambahan konsentrasi naa yaitu (0,00 mg/l, 0,10 mg/l, 0,25 mg/l, 0,50 mg/l, 0,75 mg/l, 1,00 mg/l) dan konsentrasi bap (0,00 mg/l, 0,10 mg/l, 0,25 mg/l, 0,50 mg/l, 0,75 mg/l, 1,00 mg/l). pengamatan dilakukan selama 90 hari dengan tiga parameter yaitu persentase eksplan hidup, persentase eksplan browning dan persentase eksplan kontaminasi. hasil penelitian menunjukkan persentase hidup eksplan tunas aksilar tertinggi 100%, persentase browning tertinggi 100% dan persentase kontaminasi tertinggi 66,7%. sedangkan persentase hidup daun tertinggi 100%, persentase browning tertinggi 100% dan persentase kontaminasi tertinggi 66,7%. pertumbuhan eksplan tunas aksilar dan daun stagnan hingga 90 hst, hal ini menunjukkan bahwa zat pengatur tumbuh (naa dan bap) tidak memberikan dampak apapun terhadap eksplan tunas aksilar dan daun cempaka kuning (magnolia champaca l.). kata kunci : magnolia champaca l. , kultur jaringan, tunas aksilar, daun, naa, bap.

Abstract
The effect of combination of growth regulators auxin Naphthalene Acetic Acid (NAA) with cytokinin Benzylaminopurin (BAP) on initiation of cempaka kuning (Magnolia champaca L.) axillary bud and leaves explant through in vitro, had been done. This research was conducted from January to October 2024 at the Physiology Laboratory (plant section) of the Department of Biology, Faculty of Mathematics and Natural Sciences, Syiah Kuala University. This research was designed using a completely randomized design with two factors, NAA concentration and BAP concentration. The NAA and BAP treatments consisted of 36 treatments and each treatments combination was repeated three times. The treatments used were the addition of NAA concentration (0,00 mg/l, 0,10 mg/l, 0,25 mg/l, 0,50 mg/l, 0,75 mg/l, 1,00 mg/l) and BAP concentration (0,00 mg/l, 0,10 mg/l, 0,25 mg/l, 0,50 mg/l, 0,75 mg/l, 1,00 mg/l). Data were obtained on 90 days after planting with three parameters, namely the percentage of living explants, the percentage of browning explants and the percentage of contaminated explants. The results showed that the highest percentage of living axillary bud explants was 100%, the highest percentage of browning was 100% and the highest percentage of contamination was 66.7%. While the highest percentage of living leaves was 100%, the highest percentage of browning was 100% and the highest percentage of contamination was 66.7%. The growth of axillary buds and leaves explant stagnated until 90th days, indicating that growth regulators (NAA and BAP) did not have any impact on axillary bud and leaves explants of yellow cempaka (Magnolia champaca L.).
