Abstrak permasalahan limbah di tempat pembuangan akhir (tpa) terus meningkat, terutama limbah dengan kandungan lipase. tanah yang tercemar limbah berisiko tinggi menularkan bakteri pada manusia. penelitian ini bertujuan mengidentifikasi bakteri tanah di tpa gampong jawa berdasarkan kemampuan bakteri dalam menghasilkan enzim lipase, aktivitas antibakteri, dan karakter biokimia. lima isolat bakteri (btpa3, btpa5, btpa6, btpa7, btpa16) diuji menggunakan metode corkbore untuk pengujian aktivitas lipase dan antibakteri, diikuti uji biokimia seperti mr-vp, tsia, simmons citrate, motilitas, dan indol. uji antibakteri isolat btpa5 menunjukkan zona bening 15,55 mm terhadap escherichia coli (kategori menengah) dan 8,25 mm terhadap staphylococcus aureus (kategori rendah). aktivitas lipase tertinggi ditemukan pada btpa5 dengan zona bening 31,3 mm. berdasarkan hasil identifikasi biokimia, isolat ini diduga berasal dari famili enterobacteriaceae. penelitian menyimpulkan bahwa isolat btpa5 memiliki potensi lipase tertinggi dan aktivitas antibakteri lebih efektif terhadap e. coli dibandingkan s. aureus. kata kunci : bakteri penghasil lipase, aktivitas antibakteri, tempat pembuangan akhir (tpa), gampong jawa banda aceh,, aktivitas lipase, uji antibakteri, zona bening.



Abstract

ABSTRACT The waste problem at Final Disposal Sites (FDS) continues to increase, especially waste containing lipase. Contaminated soil poses a high risk of transmitting bacteria to humans. This study aims to identify soil bacteria in FDS Gampong Jawa based on their ability to produce lipase, antibacterial activity, and biochemical characteristics. Five bacterial isolates (BTPA3, BTPA5, BTPA6, BTPA7, BTPA16) were tested using the corkbore method, followed by biochemical tests such as MR-VP, TSIA, Simmons Citrate, motility, and indole tests. Antibacterial tests on isolate BTPA5 revealed a clear zone of 15.55 mm against Escherichia coli (moderate category) and 8.25 mm against Staphylococcus aureus (low category). The highest lipase activity was observed in BTPA5 with a clear zone of 31.3 mm. Based on biochemical results, this isolate is suspected to belong to the Enterobacteriaceae family. The study concludes that BTPA5 has the highest lipase potential and antibacterial activity, which is more effective against E. coli compared to S. aureus.
