Varetas sigupai merupakan varietas lokal dari aceh bagian barat. varitas ini memiliki rasa yang enak dan beras yang harum, namun varietas ini memiliki kelemahan seperti umur tanaman dalam, arsitektur tanaman yang tinggi sehingga tanaman mudah rebah. untuk memperbaiki kelemahan tersebut dapat dilakukan dengan menginduksi gen sd-1, yang terdapat pada varietas introduksi yinzan yang memberikan hasil varietas padi dengan batang pendek. penelitian ini bertujuan untuk menganalisis keberadaan gen sd-1 dan fgr pada padi bc4f3 hasil persilangan antara varietas sigupai dan yinzhan serta melihat peluang tanaman yang prospektif untuk dilanjutkan. penelitian ini dilkasanakan di kebun percobaan dan laboratorium genetika dan pemuliaan tanaman, fakultas pertanian universitas syiah kuala sejak oktober 2022 hingga september 20023. penelitian ini menggunakan benih bc4f3 sg/yz t1.1.2.3.5, bc4f3 sg/yz t5.3.1.2., 3 bc4f3 sg/yz t5.3.12.1.3, bc4f3 sg/yz t9.1.7.6.9, bc4f3 sg/yz t9.6.1.6.12, bc4f3 sg/yz t9.8.8.3.5, bc4f3 sg/yz t29.2.11.10.7, bc4f3sg/yz t29.5.7.11.6, dan bc4f3 sg/yz t29.8.5.2.12. analisis dna menggunakan marka molekuler untuk mendeteksi adanya gen sd-1 pada turunan bc4f3 padi aceh sigupai. tanaman padi hasil persilangan sigupai dan yinzhan yang cukup prospektif untuk diteliti selanjutnya adalah bc4f3 t9.8.8.35 tanaman1 dan tanaman 5 dengan umur panen genjah berkisar antara 111-115 hss, tinggi tanaman pendek berkisar antara 89 110 cm dan hasil yang tinggi melebihi 6 ton/ha serta mewarisi gen sd-1. analisis molekuler keberadaan gen sd-1 padi bc4f3 persilangan sigupai dan yinzhan terdapat 16 tanaman secara genetik mewarisi gen sd-1 dari tetua sigupai dan yinzhan, sedangkan 62 tanaman tidak mewarisi gen sd-1 karena tidak terlihat pita dna yang sama dengan tetua sigupai dan yinzhan.



Abstract

The Sigupai variety is a local variety from the western of Aceh. This variety has a good taste and fragrant rice, but this variety has weaknesses such as the age of the inner plant, the high plant architecture so that the plant is easyly fall. To improve these weaknesses, it can be done by inducing the SD-1 gene, which is found in the Yinzan introduced variety which gives the yield of rice varieties with short stems. This study aims to analyze the existence of the SD-1 and FGR genes in BC4F3 rice from a cross between the Sigupai and Yinzhan varieties and see the opportunity for prospective plants to be continued. This research was carried out in the Experimental Gardens and Laboratory of Genetics and Plant Breeding, Faculty of Agriculture, Syiah Kuala University from October 2022 to September 2023. This study used seeds BC4F3 SG/YZ T1.1.2.3.5, BC4F3 SG/YZ T5.3.1.2., 3 BC4F3 SG/YZ T5.3.12.1.3, BC4F3 SG/YZ T9.1.7.6.9, BC4F3 SG/YZ T9.6.1.6.12, BC4F3 SG/YZ T9.8.8.3.5, BC4F3 SG/YZ T29.2.11.10.7, BC4F3SG/YZ T29.5.7.11.6, and BC4F3 SG/YZ T29.8.5.2.12. DNA analysis used molecular markers to detect the presence of the SD-1 gene in the BC4F3 derivative of Aceh Sigupai rice. Rice plants resulting from crossing Sigupai and Yinzhan which are quite prospective for further research are BC4F3 T9.8.8.35 plant 1 and plant 5. with early harvest ages ranging from 111-115 DAP, short plant height ranging from 89-110 cm and high yields exceeding 6 tons/ha and inherited the sd-1 gene. Molecular analysis of the presence of the sd-1 gene in BC4F3 rice from a cross between Sigupai and Yinzhan, there were 16 plants that genetically inherited the sd-1 gene from the Sigupai and Yinzhan parents, while 62 plants did not inherit the sd-1 gene because they did not show the same DNA band as the Sigupai and Yinzhan parents
