Abstrak tujuan dari kegiatan praktek kerja lapangan (pkl) ini adalah untuk mendeteksi bakteri salmonella sp. pada daging ayam, menggunakan media xylose lysine deoxychoalate agar (xld) dan hektoen enteric agar (hea). pemeriksaan dilakukan terhadap 15 sampel daging ayam. sampel daging ayam dikultur pada media lactose broth (lb) pada tahap pra pengayaan, dilanjutkan dengan kultur pada media tetrationate broth (ttb) dan iodine sebagai media selektif terhadap salmonella sp. pada tahap pengayaan (enrichment), dan diinkubasi selam 3 hari. suspensi selanjutnya dikultur pada media xylose lysine deoxychoalate agar (xld) dan hektoen enteric agar (hea). identifikasi keberadaan salmonella sp. dikonfirmasi dengan pengamatan karakteristik pertumbuhan salmonella sp. pada media xld dan hea. hasil pemeriksaan dengan media differensial ditemukan adanya salmonella sp. pada 6.6% (1/15) sampel. berdasarkan hasil pemeriksaan dapat disimpulkan bahwa terdapat kontaminasi salmonella sp. pada daging ayam broiler yang diperiksa di balai veteriner medan. kata kunci: daging ayam, salmonella sp., xylose lysine deoxychoalate agar, hektoen enteric agar.



Abstract

ABSTRACT The purpose of this Field Work Practice (PKL) activity is to detect Salmonella sp. bacteria in chicken meat, using Xylose Lysine Deoxychoalate Agar (XLD) and Hektoen Enteric Agar (HEA) media. The examination was conducted on 15 chicken meat samples. Chicken meat samples were cultured on Lactose Broth (LB) in pre enrichment step, and continued to Tetrationate Broth (TTB) and Iodine media as a media selektif terhadap for Salmonella sp. in enrichment step, and incubated for 3 days. The suspension was then cultured on Xylose Lysine Deoxychoalate Agar (XLD) and Hectoen Enteric Agar (HEA) media. Identification of the presence of Salmonella sp. was confirmed by inoculation on XLD and HEA media. The results of the examination with differential media found the presence of Salmonella sp. in 6.6% (1/15) of the samples examined. Based on the results of the examination, it can be concluded that there is Salmonella sp. contaminated broiler chicken meat in Balai Veteriner Medan.
