Ayam petelur (gallus gallus domesticus) merupakan ayam hasil rekayasa genetik melalui persilangan dan pemuliaan genetik secara terus-menerus. upaya budidaya ayam petelur tidak terlepas dari risiko infeksi oleh berbagai agen penyakit. salah satu penyakit yang memiliki dampak besar pada industri peternakan ayam adalah newcastle disease. untuk mengetahui status kekebalan ayam terhadap newcastle disease perlu dilakukan pengukuran titer antibodi. tujuan dari penelitian ini untuk mengetahui titer antbodi terhadap penyakit newcastle disease pada ayam layer yang telah divaksinasi. sampel yang diambil berupa serum darah dari 85 ekor ayam layer dari total populasi 400 ekor ayam yang telah divaksinasi saat usia doc, 3 pekan, dan 7 pekan. pengambilan sampel dilakukan pada pekan ke 20 di jaya m farm secara random sampling. pemeriksaan titer antibodi dilakukan dengan metode uji hemagglutination inhibition. sampel serum diinaktivasi pada suhu 56°c selama 30 menit sebelum dilakukan pengujian. antigen yang digunakan pada uji disesuaikan dengan 4 ha unit melalui uji hemagglutination assay. back titrasi dilakukan untuk memastikan titer antigen yang digunakan tepat pada 4 ha unit, sehingga didapat nilai virus optimal. hasil penelitian menunjukkan 62 dari 85 sampel serum memiliki antibodi yang protektif dengan nilai titer berkisar antara 2^4-2^8, sementara 23 sampel memiliki antibodi non-protektif terhadap newcastle disease. berdasarkan hasil penelitian dapat disimpulkan bahwa 73% ayam layer di jaya m farm memiliki titer antibodi protektif terhadap newcastle disease. kata kunci: back titrasi, hemagglutination assay, dan hemagglutination inhibition.

Abstract
Laying chickens (Gallus gallus domesticus) are chickens resulting from genetic engineering through continuous genetic crossing and breeding. Efforts to cultivate laying hens cannot be separated from the risk of infection by various disease agents. One disease that has a big impact on the chicken farming industry is Newcastle disease. To determine the immune status of chickens against Newcastle disease, it is necessary to measure antibody titers. The aim of this research was to determine the antibody titer against Newcastle disease in layer chickens that had been vaccinated. Samples taken were blood serum from 85 layer chickens from a total population of 400 chickens that had been vaccinated at DOC, 3 weeks and 7 weeks of age. Sampling was carried out in the 20th week at Jaya M Farm using random sampling. Antibody titer examination was carried out using the Hemagglutination Inhibition test method. Serum samples were inactivated at 56°C for 30 minutes before testing. The antigen used in the test is adjusted to 4 HA units through the Hemagglutination Assay test. Back titration is carried out to ensure that the antigen titer used is exactly 4 HA Units, so that optimal virus values are obtained. The results showed that 62 of the 85 serum samples had protective antibodies with titer values ranging between 2^4-2^8, while 23 samples had non-protective antibodies against Newcastle disease. Based on the research results, it can be concluded that 73% of layer chickens at Jaya M Farm have protective antibody titers against Newcastle disease.
