Concervation response unit (cru) alue kuyun dan cru sampoiniet memiliki karakteristik cuaca (suhu, kelembapan, dan curah hujan) yang berbeda. keadaan ini dapat memengaruhi kadar kortisol pada gajah sumatera di kedua cru ini. penelitian ini bertujuan untuk mengetahui kadar kortisol, suhu, kelembapan, dan curah hujan dan hubungan antar variabel pada gajah sumatera di cru alue kuyun dan cru sampoiniet. penelitian ini dilakukan terhadap 4 individu gajah sumatera (dua betina dewasa di cru alue kuyun dan 2 betina dewasa di cru sampoinet). pengukuran kortisol pada penelitian ini menggunakan kit elisa kortisol komersial eia-1887 (drg instruments gmbh, jerman) dengan total 18 sampel (delapan sampel dari cru alue kuyun dan 10 sampel dari cru sampoiniet) dan kadar kortisol diukur menggunakan metode enzyme linked-immunosorbent assay (elisa). data parameter cuaca diambil dari pusat data bmkg dan dari satelit. data perbandingan kadar kortisol dianalisis dengan mann-whitney u test dan hubungan antara kadar kortisol dengan parameter cuaca dianalisis dengan uji korelasi spearman. hasil penelitian menunjukkan kadar kortisol di cru alue kuyun dan sampoiniet tidak berbeda secara signifikan (p>0,05). berdasarkan bmkg dan satelit menunjukkan suhu di cru alue kuyun lebih rendah dibandingkan cru sampoiniet, sedangkan kelembapan dan curah hujan di cru alue kuyun lebih tinggi dibandingkan cru sampoiniet. hasil uji korelasi menunjukkan tidak terdapat hubungan yang signifikan antara kadar kortisol dengan suhu, kelembapan dan curah hujan di kedua cru tersebut. kesimpulan, kadar kortisol pada gajah sumatera di cru alue kuyun dan cru sampoiniet tidak berkorelasi dengan parameter cuaca (suhu, kelembaban, dan curah hujan).



Abstract

Alue Kuyun Conservation Response Unit (CRU) and Sampoiniet CRU have different weather characteristics (temperature, humidity and rainfall). This situation can affect cortisol levels in Sumatran elephants in these two CRUs. This study aims to determine cortisol levels, temperature, humidity, and rainfall and the relationship between variables in Sumatran elephants in Alue Kuyun CRU and Sampoiniet CRU. This study was conducted on 4 individual Sumatran elephants (two adult females in CRU Alue Kuyun and 2 adult females in CRU Sampoinet). Cortisol measurements in this study used a commercial cortisol ELISA kit EIA-1887 (DRG Instruments GmbH, Germany) with a total of 18 samples (eight samples from CRU Alue Kuyun and 10 samples from CRU Sampoiniet) and cortisol levels were measured using the enzyme linked-immunosorbent assay (ELISA) method. Data on weather parameters were taken from the BMKG data center and from satellites. Comparative data of cortisol levels were analyzed by Mann-Whitney U test and the relationship between cortisol levels and weather parameters were analyzed by Spearman correlation test. The results showed that cortisol levels in Alue Kuyun and Sampoiniet CRUs were not significantly different (P>0.05). Based on BMKG and satellite data, the temperature in Alue Kuyun CRU is lower than Sampoiniet CRU, while humidity and rainfall in Alue Kuyun CRU are higher than Sampoiniet CRU. The correlation test results showed no significant relationship between cortisol levels with temperature, humidity and rainfall in the two CRUs. In conclusion, cortisol levels in Alue Kuyun CRU and Sampoiniet CRU are not correlated with weather parameters (temperature, humidity and rainfall).
