Ikan betok (anabas testudineus) dikenal dengan nama umum climbing perch, mempunyai tekstur daging yang lembut dan gurih sehingga digemari oleh masyarakat. namun dalam perkembangannya para pembudidaya menghadapi beberapa masalah, diantaranya tidak tersedianya benih dalam jumlah yang cukup secara berkesinambungan dan ketidaksinkronan waktu kematangan gonad pada ikan jantan dan betina. salah satu cara yang dapat dilakukan yaitu melalui teknik penyimpanan sperma dan oosit. secara umum terdapat 2 jenis penyimpanan yaitu, penyimpanan beku (kriopreservasi) dan non-beku (refrigerasi). refrigerasi adalah teknik penyimpanan sel sperma atau sel telur atau material genetik lainnya pada jangka waktu pendek menggunakan suhu 0˚c-4˚c. penelitian ini bertujuan untuk mengetahui suhu preservasi yang optimum untuk oosit ikan betok berdasarkan nilai fertilisasi, daya tetas dan normalitas larva. penelitian ini menggunakan rancangan acak lengkap (ral) dengan 6 perlakuan dan 4 ulangan yaitu pa (29°c), pb (8°c), pc (4°c), pd (0°c), pe (-4°c), pf (-8°c). oosit dilakukan preservasi pada suhu perlakuan selama 120 menit, dimana setiap 30 menit oosit akan dilakukan fertilisasi. hasil penelitian preservasi oosit ikan betok menunjukkan bahwa perbedaan suhu penyimpanan berpengaruh nyata terhadap fertilitas (p0,05). persentase fertilitas tertinggi diperoleh pada perlakuan b (8°c) dimenit ke-30 dengan nilai sebesar 50,33%, nilai ini berbeda nyata pada semua perlakuan. nilai persentase daya tetas tertinggi juga diperoleh pada perlakuan b (8°c) dimenit ke-30 dengan nilai sebesar 12%, nilai ini berbeda nyata pada perlakuan e (-4°c) dan f (-8°c), namun tidak berbeda nyata dengan perlakuan a (29°c), c (4°c) dan d (0°c) (p>0,05). pengamatan pada normalitas larva yang menetas menunjukkan semua larva yang menetas berbentu normal (100%), artinya bahwa tidak ada larva abnormal yang terdeteksi. kata kunci: anabas testudineus, preservasi oosit, fertilisasi, daya tetas, normalitas larva

Abstract
Betok fish (Anabas testudineus), known as climbing perch, has a soft and tasty meat texture that is popular with the public. However, in its development, cultivators face several problems, including the unavailability of sufficient amounts of seed on an ongoing basis and the asynchronous timing of gonad maturity in male and female fish. One way that can be done is through sperm and oocyte storage techniques. In general, there are 2 types of storage: frozen storage (cryopreservation) and non-frozen (refrigeration). Refrigeration is a technique for storing sperm cells egg cells or other genetic material for a short period of time using a temperature of 0˚C-4˚C. This study aims to determine the optimum preservation temperature for betok fish oocytes based on fertilization value, hatchability and larval normality. This study used a completely randomized design (CRD) with 6 treatments and 4 replications, namely PA (29°C), PB (8°C), PC (4°C), PD (0°C), PE (-4°C), PF (-8°C). Oocytes were preserved at treatment temperature for 120 minutes, where every 30 minutes oocytes would be fertilized. The results of research on the preservation of betok fish oocytes showed that differences in storage temperature had a significant effect on fertility (P0.05). The highest percentage of fertility was obtained in treatment B (8°C) in the 30th minute with a value of 50.33%, this value was significantly different in all treatments. The highest hatchability percentage value was also obtained in treatment B (8°C) in the 30th minute with a value of 12%, this value was significantly different in treatment E (-4°C) and F (-8°C), but not significantly different significantly with treatments A (29°C), C (4°C) and D (0°C) (P>0.05). Observations on the normality of hatched larvae showed that all hatched larvae were normal (100%), meaning that no abnormal larvae were detected.
