Penelitian yang berjudul studi filogenetik tumbuhan salak (salacca sp.) di daerah aceh berdasarkan cpdna trnl-f intergenic spacer ini bertujuan untuk menganalisis dan merekonstruksi hubungan kekerabatan tumbuhan salak budidaya dan liar asal aceh melalui analisis filogenetik menggunakan trnl-f intergenic spacer. penelitian ini dilakukan dari bulan juli 2019 hingga maret 2020. data sekuen dna dianalisis hubungan kekerabatannya dengan cara disejajarkan pada clustalw multiple alignment dalam software ugene. analisis rekonstruksi pohon filogenetik menggunakan program paup * versi 4.0b10 berdasarkan metode maximum parsimony (mp) dan neighbor joining (nj). pengambilan sampel salak diperoleh dari beberapa lokasi berdasarkan informasi dari masyarakat setempat yaitu lembah seulawah, sabang, montasik, kutacane, dan kawasan ekosistem leuser. rekonstruksi pohon filogenetik yang dihasilkan menunjukkan bahwa sampel salak terbagi dalam dua cabang dan mencirikan kelompok jenis salak yang berbeda. cabang i menunjukkan bahwa kelompok ini terdiri dari salak budidaya. cabang ii menunjukkan bahwa percabangan ini mengarah pada kelompok salak liar. analisis mp menunjukkan bahwa nh.05 (lembah seulawah) dan nh.ma (kutacane) merupakan jenis salak yang paling modern, sedangkan sampel nh.r3 (kawasan ekosistem leuser) yang paling primitif. analisis nj juga menunjukkan bahwa salak budidaya lebih modern dibandingkan salak liar. kata kunci : filogenetik, salacca sp., aceh, trnl-f intergenic spacer



Abstract

The study of plant phylogenetic Salak (Salacca sp.) in Aceh area based on trnL-F intergenic spacer, aimed to analyzed and reconstructed the relationship between cultivated and wild Salacca from Aceh through phylogenetic analysis using trnL-F intergenic spacer. The study was conducted from July 2019 until March 2020. DNA sequence data were analyzed for their kinship relationships by alignment them with ClustalW Multiple Alignment in UGENE software. The phylogenetic tree reconstruction analysis used PAUP * version 4.0b10 program based on the Maximum Parsimony (MP) and Neighbor Joining (NJ) methods. Salacca samples were obtained at several locations based on information from local communities were Lembah Seulawah, Sabang, Montasik, Kutacane, and Kawasan Ekosistem Leuser. The reconstruction of phylogenetic trees that were produced showed that the Salacca samples were divided into two branches and characterized the different groups of Salacca species. Branch I showed that this group consisted of cultivated Salacca. Branch II showed that this branching leaded to the wild Salacca. Maximum Parsimony analysis showed that sample NH.05 (Lembah Seulawah) and sample NH.MA (Kutacane) were most modern, while sample NH.R3 (Kawasan Ekosistem Leuser) was most primitive. Neighbor Joining analysis also showed that cultivated group was more modern that wild group. Keywords : Phylogenetic, Salacca sp., Aceh, trnL-F intergenic spacer
