PENGARUH PEMBERIAN PGF2? DALAM PENGENCER
SEMEN TERHADAP PENINGKATAN POST THAWING
MOTILITY SPERMATOZOA DOMBA WARINGIN

ABSTRAK
Prostaglandin (PGF2?) merupakan salah satu hormon yang berperan penting dalam sistem reproduksi. Tujuan dari penelitian ini adalah mengetahui pengaruh pemberian PGF2? dalam pengencer semen komersial (Andromed) terhadap peningkatan PTM spermatozoa domba Waringin. Dalam penelitian ini digunakan sampel semen dari 3 ekor domba Waringin berumur 2 - 3 tahun yang dikoleksi menggunakan vagina buatan dan kualitasnya dievaluasi secara makroskopis dan mikroskopis. Sampel semen yang telah dievaluasi ditambah pengencer Andromed, kemudian dibagi atas 3 kelompok perlakuan, yaitu: P1, P2, dan P3 yang masing-masing ditambahkan NaCl 0,9%, 37,5 µg PGF2?, dan 75 µg PGF2?. Seluruh sampel disimpan dalam refrigerator selama 4 jam dan dilakukan pemeriksaan motilitas spermatozoa, kemudian dilakukan proses pembekuan (freezing) selama 24 jam dan dilakukan pemeriksaan motilitas post thawing spermatozoa. Data yang diperoleh dianalisis menggunakan analisis ragam pola satu arah (ANOVA) dan dilanjutkan dengan uji Duncan. Hasil penelitian menunjukkan, bahwa motilitas pre-freezing spermatozoa (%) pada P1, P2, dan P3 masing-masing adalah 43,67 ± 1,20; 75,00 ± 4,00; dan 82,33 ± 3,18; dan motilitas spermatozoa post thawing (%) pada P1, P2, dan P3 masing-masing adalah 25,33 ± 1,85; 49,33 ± 3,17; dan 54,00 ± 5,50. Disimpulkan bahwa penambahan PGF2? pada pengencer Andromed dapat meningkatkan motilitas spermatozoa domba Waringin.

THE IMPACT OF PGF2? ADMINISTRATION IN SEMEN DILUENTS
ON THE INCREASE OF POST THAWING SPERM MOTILITY
IN WARINGIN RAM

ABSTRACT
Prostaglandin (PGF2?) is an important hormone in the reproductive system. The aim of the study was to determine the effect of PGF2? in commercial semen diluents (Andromed) on the improvement of Waringin ram sperm motility. This study used semen from three Waringin rams aged 2 - 3 years old collected using artificial vagina. The quality of sperm evaluated macroscopically and microscopically. After evaluated, semen samples were added with Andromed diluents then divided into three group treatments (P1, P2, and P3) where each group was then added with 0.9% NaCl, 37.5 µg PGF2?, and 75 µg PGF2?. All of the samples were stored in refrigerator for 4 hours and sperm motility was examined. The freezing process was carried out for 24 hours and subsequently the post thawing sperm motility was examined. The data obtained were analyzed by one-way analysis of variance (ANOVA) follow the Duncan test. The result showed that motality of pre-freezing sperm (%) of Waringin ram at P1, P2, and P3 treatments were 43.67 ± 1.20, 75.00 ± 4.00, and 82.33 ± 3.18 respectively. The motility of post thawing sperm (%) of Waringin rams at P1, P2, and P3 treatments were 25.33 ± 1.85, 49.33 ± 3.17, and 54.00 ± 5.50 respectively. The conclusion of this study is the addition of PGF2? to Andromed diluents can increase the motility of Waringin rams.
