Antibiotik merupakan bahan yang dikeluarkan oleh mikroorganisme dan bersifat antagonis terhadap pertumbuhan dan hidupnya mikroorganisme lain. Penggunaannya yang luas menyebabkan bakteri resisten terhadap antibiotik tertentu. Penelitian ini bertujuan untuk melihat kerentanan Salmonella sp. asal kura-kura ambon (Cuora amboinensis) terhadap beberapa jenis antibiotik. Koleksi sampel diambil dari stok bakteri Salmonella sp. yang diisolasi dari kura-kura ambon di Laboratorium Mikrobiologi FKH Unsyiah. Penelitian ini dilakukan dengan metode difusi cakram menurut Kirby-Bauer. Koleksi bakteri dikultur ulang dengan melakukan penanaman ke dalam Nutrient Broth (NB), lalu, diinkubasi pada suhu 37oC selama 24 jam dan diamati kekeruhannya sesuai dengan standar larutan Mc. Farland 0,5. Bakteri ditanam pada Mueller Hinton Agar (MHA), kemudian, cakram antibiotik siproflaksin, gentamisin, tetrasiklin, amoksisilin, kloramfenikol dan streptomisin, diambil dan diletakkan di permukaan Mueller Hinton Agar (MHA) yang telah diolesi bakteri tersebut. Media Mueller Hinton Agar (MHA), diinkubasi pada temperatur 370C selama 24 jam. Sensitivitas masing-masing antibiotik dicatat berdasarkan diameter zona hambatan yang terbentuk disekitar cakram, dan dibandingkan dengan tabel Clinical and Laboratory Standars Institute (CLSI). Data disajikan dalam bentuk tabel dan gambar dan dianalisis secara deskriptif. Hasil penelitian menunjukkan bahwa pada lima belas isolat Salmonella sp. asal kura-kura ambon telah resisten terhadap antibiotik tetrasiklin (100%), amoksilin (100%), kloramfenikol (100%) dan streptomisin (80%) serta bersifat intermediet terhadap antibiotik gentamisin (40%) dan siprofloksasin (14%). Hasil penelitian juga menunjukkan bahwa isolat Salmonella sp., masih sensitif terhadap antibiotik siprofloksasin (86%) dan gentamisin (60%).

Antibiotics are a substances released by microorganisms and act as antagonist agent against the growth and life of other microorganisms. Extensive use of antibiotics, without a proper consideration of its rational use, may develop a resistance against a specific antibiotic. This research aims to determine the susceptibility of Salmonella sp. isolated from ambon turtles (Cuora amboinensis) against several types of antibiotics. A total of 15 Salmonella sp. isolate which were provided by Microbiology Laboratory of Faculty of Veterinary Medicine, Universitas Syiah Kuala were used in this study. This research was conducted using the Kirby-Bauer disc diffusion method. The bacterial collection was re-cultured by inoculating into Nutrient Broth (NB), then, incubated at 37oC for 24 hours. The turbidity of bacteria were observed by comparing with 0,5 Mc. Farland standard. The bacteria were grown on Mueller Hinton Agar (MHA) and the antibiotics disc containing ciprofloxacin, gentamicin, tetracycline, amoxicillin, chloramphenicol and streptomycin were placed, on the surface of Mueller Hinton Agar (MHA), then incubated at 37o C for 24 hours. The activity of each antibiotic was carried out based on the diameter of inhibition zone that formed around the disc and compared with the Clinical and Laboratory Standards Institude (CLSI). The data were tabulated and analysed descriptively. The results showed that fifteen Salmonella sp. isolated from ambon turtles (Cuora amboinensis) were resistant to tetracyclines (100%), amoxicillin (100%), chloramphenicol (100%) and streptomycin (80%) and intermediet to gentamicin (40%) and ciprofloxacin (14%). However, the isolates were still sensitive to gentamicin (60%) and ciprofloxacin (86%).
