Enterococcus faecalis merupakan jenis bakteri patogen oportunistik yang dapat ditemukan pada saluran akar gigi. Kandungan senyawa antibakteri yang terkandung dalam tanaman nilam Aceh (Pogostemon cablin Benth.) berpotensi sebagai alternatif dalam penghambatan pertumbuhan Enterococcus faecalis. Penelitian ini bertujuan untuk mengetahui efisiensi ekstraksi daun nilam Aceh (Pogostemon cablin Benth.) dan mengetahui daya hambat, serta nilai Minimum Inhibitory Concentration (MIC) ekstrak etanol daun nilam Aceh terhadap bakteri Enterococcus faecalis. Penelitian ini menggunakan metode maserasi dalam pembuatan ekstrak. Pengukuran diameter zona hambat menggunakan metode Kirby Bauer untuk menguji aktivitas antibakteri yang dilakukan dengan perlakuan konsentrasi ekstrak 1,56%, 3,12%, 6,25%, 12,5%, 25%, kontrol positif menggunakan klorheksidin (CHX 0,2%) dan kontrol negatif menggunakan akuades steril. Penentuan Minimum Inhibitory Concentration (MIC) dilakukan menggunakan spektrofotometri UV-Vis dengan pengukuran nilai Optical Density (OD) pada panjang gelombang 610 nm. Hasil penelitian didapatkan nilai rendemen total ekstrak etanol daun nilam Aceh sebesar 17,25% dan memiliki aktivitas antibakteri terhadap Enterococcus faecalis. Analisis zona hambat pada perlakuan konsentrasi 25% memiliki aktivitas daya hambat sedang, sedangkan pada perlakuan konsentrasi 12,5%, 6,25%, 3,12% dan 1,56% masih memiliki aktivitas daya hambat yang lemah. Nilai Minimum Inhibitory Concentration (MIC) yang diperoleh berada pada konsentrasi ekstrak 12,5%.

Enterococcus faecalis is a type of opportunistic pathogenic bacteria that can be found in the root canals of teeth. The content of antibacterial compounds contained in Aceh patchouli (Pogostemon cablin Benth.) has the potential as an alternative in inhibiting the growth of Enterococcus faecalis. This study aims to determine the extraction efficiency of Aceh patchouli leaves (Pogostemon cablin Benth.) and to determine the inhibitory power and Minimum Inhibitory Concentration (MIC) value of the ethanol extract of Aceh patchouli leaves against Enterococcus faecalis bacteria. This study used the maceration method in making extracts. Measurement of the diameter of the inhibition zone used the Kirby Bauer method to test antibacterial activity which was carried out by treating extract concentrations of 1.56%, 3.12%, 6.25%, 12.5%, 25%, positive control using chlorhexidine (CHX 0.2 %) and negative control using sterile distilled water. Determination of Minimum Inhibitory Concentration (MIC) was carried out using UV-Vis spectrophotometry by measuring the Optical Density (OD) value at a wavelength of 610 nm. The results showed that the total yield value of the ethanol extract of Aceh patchouli leaves was 17.25% and had antibacterial activity against Enterococcus faecalis. Inhibition zone analysis at 25% concentration had moderate inhibitory activity, while at 12.5%, 6.25%, 3.12% and 1.56% concentrations still had weak inhibitory activity. The Minimum Inhibitory Concentration (MIC) value obtained was at an extract concentration of 12.5%.
