Semen is a fluid secreted by the male reproductive organs and is typically ejaculated into the female reproductive tract during copulation. Assessing semen quality is an important tool for predicting the reproductive performance of male cattle. The quality of chicken semen is influenced by various factors, including the chicken breed. This study aimed to compare the macroscopic and microscopic characteristics of semen quality among country chickens, bantam chickens, and red jungle fowl. A completely randomized design (CRD) was employed in this study, using local chicken breeds with three replications. The results revealed that the semen volume of country chickens was approximately 0.28 ± 0.03 mL/ejaculate, which was higher than that of bantam chickens (0.17 ± 0.06 mL/ejaculate) and red jungle fowl (0.15 ± 0.05 mL/ejaculate). The pH of country chicken semen was approximately 7.33 ± 0.31, similar to that of bantam chicken semen, while red jungle fowl semen had a pH of about 7.20 ± 0.20. The spermatozoa concentration in country chickens was approximately 2260.00 ± 104.40 million/mL, which was higher than that of bantam chickens (1980.00 ± 157.16 million/mL), while red jungle fowl had a concentration of about 1736.67 ± 317.86 million/mL. The motility of country chicken spermatozoa was approximately 85.11 ± 3.29%, bantam chicken had a motility of about 80.43 ± 1.82%, and red jungle fowl exhibited a motility of around 82.05 ± 2.83%. The spermatozoa viability in country chickens was approximately 87.61 ± 2.94%, bantam chicken had a viability of about 85.05 ± 2.63%, and red jungle fowl had a viability of approximately 86.91 ± 1.84%. Regarding abnormal spermatozoa, country chickens had a percentage of about 6.63 ± 0.95%, bantam chickens had 6.79 ± 1.52%, and red jungle fowl exhibited 7.58 ± 1.43%. Based on the study's findings, it can be concluded that the semen volume and spermatozoa concentration of country chickens were significantly higher (P0.05) observed in the pH, color, consistency, motility, viability, and abnormal spermatozoa characteristics among country chickens, bantam chickens, and red jungle fowl.
