PADA JARINGAN TESTIS SAPI ACEH MENGGUNAKAN
TEKNIK IMUNOHISTOKIMIA


ABSTRAK
Protein alpha smooth muscle actin (α-SMA) memiliki fungsi kontraktil yang ditemukan
pada sel-sel otot polos pada berbagai jaringan tubuh seperti testis. Pada testis α-SMA
diekspresikan oleh sel-sel myoid peritubular dan otot polos, buluh darah. Penelitian ini bertujuan
mengetahui sebaran dan ekspresi -SMA pada testis sapi aceh yang dideteksi dengan teknik
imunohistokimia (IHK). Penelitian ini menggunakan lima pasang organ testis sapi aceh jantan
dewasa berumur 2-3 tahun. Sampel testis difiksasi dengan larutan neutral buffer formalin (NBF)
10 %, diproses menjadi preparat histologi lalu diwarnai dengan pewarnaan IHK metode avidin
biotin complex peroxidase (ABC method). Sebaran α-SMA dianalisis secara deskriptif sedangkan
ekspresi α-SMA dinilai dengan pemberian skor intensitas kemudian hasilnya dianalisis
menggunakan uji Kruskal-Wallis dan uji lanjut Mann Whitney U Test. Hasil penelitian
menunjukkan bahwa α-SMA ditemukan pada sel myoid peritubular, buluh darah, jaringan ikat,
dan serabut otot polos tunika albuginea testis. Berdasarkan uji statistik terdapat perbedaan nyata
ekspresi α-SMA pada jaringan ikat dengan sel myoid peritubular, buluh darah, dan serabut otot
polos (P

DETECTION AND DISTRIBUTION OF ALPHA SMOOTH MUSCLE ACTIN IN TESTICULAR TISSUE OF ACEH CATTLE USING IMMUNOHISTOCHEMICAL TECHNIQUES ABSTRACT Alpha smooth muscle actin protein (α-SMA) has contractility function that found in smooth muscle cells in various body tissues such as in the testicular tissue. The protein α-SMA is specifically expressed by peritubular myoid cells and blood vessel smooth muscle cells. This study aims to determine the distribution and expression of α-SMA in the testes of aceh cattle detected by immunohistochemical (IHC) techniques. Five pairs of testes from five adults aceh cattle aged 2-3 years were used in this study. All testicular samples were fixed using 10% neutral buffer formalin (NBF 10%) solution and then processed into histological preparations and stained with IHC staining using avidin biotin complex peroxidase (ABC method) method. The distribution of αSMA was analyzed descriptively where as the expression of α-SMA was evaluated by giving an intensity score and then analyzed using the Kruskal-Wallis test and followedby Mann Whitney U Test. The results showed that α-SMA was found in peritubular myoid cells, blood vessels, connective tissue, and smooth muscle fibers of the tunica albuginea testis.
