Abstract
Background: Honey has been widely used as a wound healing therapy in various parts of the world, including Aceh, Indonesia, due to its antibacterial and antiinflammatory properties. However, limited research on the formulation of honey ingredients in the form of modern dressings is available. Objective: To assess the effect of Aceh kelulut honey-based hydrocolloid dressing on wound healing macroscopically and microscopically. Methods: 30 healthy adult male mice, 2-3 months old, were given two open wounds on their backs with a diameter of 4 mm. The mice were then divided into three groups: one group received normal saline without Aceh kelulut honey (K), one group received pure Aceh kelulut honey (MK) and one group received Aceh kelulut honey-based hydrocolloid dressing (HC). Wound diameter was calculated daily until day 14. Skin biopsies were taken and examined histopathologically on days 3, 7, and 14 after treatment. Histological assessment of wound healing was based on the number of neutrophils, macrophages, epithelialization ratio, myofibroblasts, and collagen fiber density. Data were analyzed using ANOVA or Kruskal-Wallis statistical test, followed by Mann Whitney test. Results: Statistical test results showed that there were significant differences in the number of neutrophils, epithelialization ratio, myofibroblast ratio, and collagen fiber density ratio between the HC, MK and K groups (p0.05). Conclusion: The application of Aceh kelulut honey-based hydrocolloid dressing to the wound can accelerate wound healing, reduce the number of neutrophils, increase the ratio of myofibroblasts, and increase the density ratio of collagen fibers, but inhibit the ratio of wound epithelialization and do not affect the number of macrophages in both the inflammatory phase and the proliferation phase. Keywords: hydrocolloid dressing, Aceh kelulut honey, neutrophil, macrophages, epithelialization
