Abstrak penelitian ini bertujuan untuk mengetahui pengaruh pemberian ekstrak biji putat air (barringtonia racemosa) terhadap jumlah eritrosit, kadar hemoglobin, dan persentase hematokrit tikus putih (rattus norvegicus) yang dipapar asap rokok. sebanyak 25 ekor tikus putih (rattus norvegicus) strain wistar, usia 3-4 bulan, berjenis kelamin jantan dengan bobot 180-200 gram, secara acak dibagi menjadi 5 kelompok perlakuan, masing-masing kelompok terdiri dari 5 ekor tikus. kelompok kontrol negatif (kn) tanpa dipaparkan asap rokok dan tanpa pemberian ekstrak putat air (barringtonia racemosa), kelompok kontrol positif (kp) dipaparkan asap rokok tanpa pemberian ekstrak putat air (barringtonia racemosa), kelompok perlakuan p1, p2, p3 dipaparkan asap rokok dan diberi biji putat air (barringtonia racemosa) dengan dosis berturut 50mg/kgbb/hari, 100 mg/kgbb/hari, dan 150 mg/kgbb/hari. ekstrak diberikan secara oral selama 30 hari. pengambilan darah pada semua kelompok melalui vena orbitalis dengan pipet hematokrit setelah disedasi dengan 0,2 ml ketamin. kemudian nilai eritrosit, hemoglobin, dan hematokrit dihitung dengan metode pemeriksaan hematologi menggunakan hematology analyzer. rata-rata (±sd) kn, kp, pi, p2, dan p3 nilai eritrosit (10 /µl) berturut-turut adalah 7,17±1,16; 5,06±1,33; 7,21±1,08; 7,33±1,18; 6,73± 0,93; hemoglobin (g/dl) berturut-turut adalah 16,00±4,12; 11,20±4,04; 16,16±2,55; 17,34±2,82; 15,70±0,81, hematokrit (%) berturut-turut adalah 41,29±6,20; 32,24±7,57; 42,04±4,84; 45,38±5,34; 40,80±3,87. hasil penelitian menunjukkan bahwa pemberian biji putat air (barringtonia racemosa) pada dosis 100mg/kgbb/hari berpengaruh nyata dalam meningkatkan nilai dari eritrosit, hemoglobin, dan hematokrit dibandingkan ekstrak putat air dosis 50mg/kgbb/hari dan 150mg/kgbb/hari. kesimpulan pemberian ekstrak putat air dapat meningkatkan jumlah eritrosit, kadar hemoglobin, dan persentase hematokrit yang dipaparkan asap rokok. kata kunci: asap rokok, putat air, eritrosit, hemoglobin, hematokrit abstrac this study aims to determine the effect of giving water putat seed extract (barringtonia racemosa) on the number of erythrocytes, hemoglobin levels, and hematocrit percentage of white rats (rattus norvegicus) exposed to cigarette smoke. a total of 25 white rats (rattus norvegicus) wistar strain, aged 3-4 months, male, weighing 180-200 grams, were randomly divided into 5 treatment groups, each group consisting of 5 rats. the negative control group (kn) without being exposed to cigarette smoke and without giving water putus extract (barringtonia racemosa), the positive control group (kp) were exposed to cigarette smoke without giving hipster extract (barringtonia racemosa), the treatment groups p1, p2, p3 were exposed to cigarette smoke and given water putat seeds (barringtonia racemosa) in successive doses of 50 mg/kg/days, 100 mg/kg/days, and 150 mg/kg/days. the extract was administered orally for 30 days. blood was drawn from all groups through the orbital vein with a hematocrit pipette after sedation with 0.2 ml of ketamine. then the erythrocyte, hemoglobin, and hematocrit values were calculated by the hematological examination method using a hematology analyzer. the mean kn, kp, pi, p2, dan p3 (±sd) erythrocyte values (106 / µl) were 7.17 ± 1.16, respectively; 5.06 ± 1.33; 7.21 ± 1.08; 7.33 ± 1.18; 6.73 ± 0.93; hemoglobin (g / dl) were 16.00 ± 4.12, respectively; 11.20 ± 4.04; 16.16 ± 2.55; 17.34 ± 2.82; 15.70 ± 0.81, the hematocrit (%) were 41.29 ± 6.20, respectively; 32.24 ± 7.57; 42.04 ± 4.84; 45.38 ± 5.34; 40.80 ± 3.87. the results showed that giving water putat seeds (barringtonia racemosa) at a dose of 100mg/kgbb/day had a significant effect on increasing the value of erythrocytes, hemoglobin, and hematocrit compared to water putat extract at a dose of 50mg/kgbb/day and 150mg/kgbb/day. the conclusion is that giving water spinach extract can increase the number of erythrocytes, hemoglobin levels, and the percentage of hematocrit exposed to cigarette smoke. keywords: cigarette smoke, putat air, eryrocytes, hemoglobin, hematocrit
