Abstrak domba ekor gemuk merupakan salah satu jenis domba yang banyak dikembangbiakkan sebagai penghasil daging. keempukan pada daging dapat dipengaruhi oleh beberapa faktor seperti pakan, lingkungan dan gen. gen kalpain-1 (capn1) menghasilkan enzim kalpain yang dapat mengontrol struktur protein dan keempukan daging. pengetahuan mengenai marker gen dapat membantu upaya pengembangbiakan serta seleksi domba, hasil dari marker gen dapat membantu peternak dalam mengembangbiakkan domba ekor gemuk unggul. tujuan penelitian ini untuk mengetahui keragaman gen kalpain-1 (capn1) pada domba ekor gemuk di kabupaten bireuen. penelitian menggunakan metode polymerase chain reaction-restriction fragment length polymorphism (pcr-rflp) yang kemudian dianalisis menggunakan hukum hardy-weinberg. hasil penelitian menunjukkan gen kalpain-1 (capn1) teramplifikasi pada ukuran ~320 pb. berdasarkan hasil rflp fragmen deoxyribose nucleic acid (dna) diamplifikasi menjadi dua bagian yang teletak pada ukuran ~210 pb dan ~110 pb. hal ini mengindikasikan bahwa keragaman gen kalpain-1 (capn1) pada domba ekor gemuk tidak terdapat mutasi dan bersifat monomorfik. kata kunci: kalpain-1, gen, monomorfik, pcr-rflp

Abstract
ABSTRACT Fat tail sheep is one type of sheep that is widely bred as a meat producer. Tenderness in meat can be influenced by several factors such as feed, environment, and genes. The calpain-1 (CAPN1) gene produces calpain enzymes that can control the protein structure and tenderness of the meat. Knowledge of gene markers can help the breeding and selection of sheep, and the results of gene markers can help breeders in breeding superior fat tail sheep. The purpose of this study was to determine the diversity of the calpain-1 (CAPN1) gene in fat-tailed sheep in Bireuen Regency. The study used the polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method which was then analyzed using the Hardy-Weinberg law. The results showed that the calpain-1 (CAPN1) gene was amplified at ~320 bp. Based on the results of the RFLP, the deoxyribose nucleic acid (DNA) fragment was amplified into two parts that were located at a size of ~210 bp and ~110 bp. This indicates that the diversity of the calpain-1 (CAPN1) gene in fat-tailed sheep has no mutations and is monomorphic. Keywords: Calpain-1, gene, monomorphic, PCR-RFLP
